/cnick is handy for changing nick colours without using the addressbook dialog.

Is there a similar command for changing highlighting parameters on the fly?

Not in the niclist. Use a dll like Nicklust if you want that.

There isn't (you might want to suggest that).

The closest thing you can do is put variables in the Highlight section and modify those.

Apart from normal text, an entry in Highlight can be a variable. So you can add "%myvar" there, then whenever you want to modify the entry, you just do a /set %myvar new-value-here.

This is far from equivalent to a /highlight command of course (which presumably would also allow you to add items as well as modify the various properties, like colour or message/nickname trigger)
